Sujet : Fluorescence microscopy is a widely-used imaging modality given its strong signal, selective labelling within complex systems and compatibility with super-resolution methods reaching sub-diffraction scale spatial resolution (~20nm). This technique allows accessing the orientation of molecules, making it a unique tool for studying structural properties at the nanoscale and understanding new phenomena in nanophysics. Recently, the MOSAIC team at has brought pioneering contributions to quantify both the 3D localization and 3D orientation of single fluorescent molecules in biological samples. These methods exploit polarization projections combined with either PSF-engineering techniques or pupil splitting.
In this internship, the student will participate to the ongoing progress to push further the performances of the existing approaches, and to address new challenges. The work can be either fully experimental, fully computational or a mix of both. It will consist in participating in the current experimental and/or computational efforts, addressing one of these two topics : (1) to improve the performance of the codes currently developed to analyse the data and expend them with new experimental capabilities such as real time orientation detection ; (2) to address novel experimental challenges such as accounting for optical aberrations in a polarized microscope, tracking single molecules’ orientations in times, or enlarging the depth extent of super resolution polarized microscopy.
