"Real-time dynamics of molecular order and orientation by fast polarization modulation in coherent Raman scattering microscopy"

Hamamatsu Photonics Best Presentation Award" of the SPIE Photonics West 2019 BiOS conference (2-7 February, San Francisco, USA)

Title of talk: "Real-time dynamics of molecular order and orientation by fast polarization modulation in coherent Raman scattering microscopy"

Content:
The sensitivity of resonant vibrational modes to the incident polarization in Coherent Raman Scattering (CRS) gives additional insight into the structural order and orientation of molecular assemblies. The limiting factor of polarized-CRS imaging for dynamic biological imaging is however its long acquisition times. Here we show how to overcome this limitation by fast electro-optic modulation of the polarization state, allowing up to sub-seconds time scales per image. Coupled with lock-in detection, we demonstrate real-time imaging of the deformation and dynamics of model membrane systems (multilamellar vesicles) and single lipid bilayer sensitivity as present in red blood cell ghosts.
